These primers were designed to amplify unique (non-cross-hybridizing) fragments within yeast ORFs that are otherwise predicted to cross-hybridize.
They were generated with
Primer 3
(Skaletsky & Rozen, Whitehead Institute) and the following
rules
from Pete Estep: G + C = 12 bases, 20 to 23 bases total. In addition,
the fragment length was maximized and the difference in Tm between the primers minimized.
